聚乳酸载药微球制备及释药性能研究最新进展

摘要:

对可生物降解材料聚乳酸作为药物载体制备微球制剂的研究状况进行了综述。针对目前限制聚乳酸微球制剂临床应用存在的问题,重点介绍了降低药物突释,提高药物包封率,改善多肽和蛋白药物微球释药性能等方面研究的最新进展。聚乳酸载药微球在药物传输中有着广阔的研究和应用前景。

Abstract:

Poly(L-lactide) (PLLA) is a biodegradable and biocompatible semi-crystalline polymer, with resultant environmental advantages among synthetic polymers. It can be applied as controlled-release devices, absorbable sutures, implants for orthopaedic use, absorbable fibres and disposable degradable plastic. The researches of preparation and drug release characteristics of PLA microspheres for the drug delivery system are summarized. It focuses on the recent developments in resolving the existent problem of clinical application, ranging from reduce initial burst of drug release, increase encapsulation efficiency to improve the drug release characteristics of protein microsphere. The multi-reservoir type microspheres composed of PLGA and PLA is one way to efficiently reduce initial burst. The chemical modification of PLA is another way to increase encapsulation efficiency and improve the drug release characteristics. The review mainly aims to promote the investigation of PLA microspheres for worldwide researcher.
